I'm not sure about the speed, because there is a writing and reading speed.

I have a Lexar 16GB 400x and the recording stops after 3 seconds

It will work but 5D2 will not take full advantage of this card's performance. Write speed benchmarks are not even close to 100 MByte/s.
www.cameramemoryspeed.com has some reliable benchmark numbers (for cards in cardreaders).
